Expression of HIV-1 antisense transcript during viral infection. (A) 293T cells were infected with 35 ng of NL4.3LucR-E- or NL4.3AsLucR-E- virions pseudotyped
with Vesicular Stomatitis Virus envelope (VSVg) and were lysed at different time points
post-infection and tested for luciferase activity. In (B), 10 μM or 50 μM of AZT was added to the culture media 1 h before infection of 293T
cells and luciferase activity was determined after 48 hr. (C) SupT1, Jurkat, CEMT4 and U937 cells were infected with NL4.3LucR-E- or VSVg-pseudotyped
NL4.3AsLucR-E- virions at a ratio of 1 ng/1 × 105 cells. Cells were lysed 48 hr post-infection and tested for luciferase activity. (D) Forty-eight hr post-infection, Jurkat cells were seeded at a concentration of 1
× 106 cells/ml and stimulated with various cell activators for 8 h. Luciferase activities
represent the mean value of three measured samples ± S.D. Luciferase activity in panels
A, B and C are presented on a logarithmic scale. Mean values of luciferase activity are indicated
in panels A and C above each column.
